YOU NEED TO IMAGE AN OBJECT OF A KNOWN LENGTH, SUCH AS A RULER OR PRECISION SPHERES.

How do you calculate for this image, can you please tell me an code ? here is the my image attached, Please do let me know the matlab code to calculate further dimension of any other images. Thank you so much

Devesh, I did tell you and did give you the code. Why didn't you run my attached demo and draw a line across your micron bar on the bottom to calibrate, and then draw the vertical distance to get the height of the object?
Or to get automatically, do your spatial calibration and then threshold the image and get the bounding box, and then multiply the bounding box height in pixels by your micronsPerPixel calibration factor. See my Image Segmentation Tutorial in my File Exchange for a full demo:
