Measuring diameter of blood vessels

Hello,
I have a project that I would like to understand how to approach. I'm given a task to measure the diameter of a blood vessel, I think Matlab would be the best for image processing because I have most experience with it. I'm sure there are multiple libraries already existing that can do such a task, I would just like some help understand the methods behind it.
