What is the use of plot(x, y, ':') while plotting the graph?

x=0:pi/100:2*pi;
y=cos(x);
y2=sinx
plo(x,y2, ':')
why is ':' required?

But plot(x,y2) also shows the results.

Did you perhaps mean plot(x,y2,':')?
If so, read the documentation for the plot function. It will clearly explain what that does.

':' is dotted line joining the points.
