How to check the color variation in the ROI of an image

Hi All,
I have many coloured images,please could anyone tell me how to check the color variation in the ROI (segmented image) of an image?

One reasonable way would be to calculate the mean color of the image. Then calculate the delta E (color difference) from every pixel to that mean color. Then histogram the delta E image. See http://www.mathworks.com/matlabcentral/fileexchange/31118-color-segmentation-by-delta-e-color-difference
